 Not sure about Mame32 but I can successfully launch Mame Plus
from a slaved Hard drive. I can only assume it would be the same?

Yes you can even run it from a network share.

sure. most of my PC's have a small main drive and a large slave.
copy short cut to desktop or startup folder.

you can even have it on CD od DVD

there are some CD's around that boots at startup but I have never had luck with em. (mame boot disks)
there just more work than I care to put in them.

 What version of Mame, are you using, on your flash drive?
 Would you have the proceedure, on how to load Mame to your flash drive, and launch the program?
I have thumb drive with mame32 and games on it and it plays fine

Just copy mame32 to the thumb drive and load any games you want to the correct directory.  Insert the thumb drive, launch mame32.exe, find the game you want to play that you copied to the directory and play away.  I use version 113 since that is what my rom set is
