It sounds like you might have some wiring issues with P3 & P4 controls (if two player games are OK). Check P3 & P4 microswitches and confirm that there are no NC wiring stuff-ups.

How did you configure the mame inputs - in game hitting Tab and then into either Input (general) or (this game)?  The default inputs in mame are not much help for those with 4-player cabs as not all controls are mapped and some of P3 controls are linked to P2 controls.

www.mameworld.net/easyemu - this will help if you want to set up a ctrl file which sets out which controls you want to use (can make it game specific as well)
