Hi! If I want to use Gsync with Mame must I enable vsync in the Mame options?

No.
vsync is for people with displays that are hardlocked to a specific hZ and then you get nasty side effects like tearing or audio screw-ups.
By having a G-Sync capable display and video card you eliminate having to use vsync
