Take multiple songs and mix them to make a new thing. A Night at the Hip-Hopera by the Kleptones (Rap songs mixed with Queen) is a favorite of mine. The Gray Album is a famous example by DJ Danger Mouse.

Well, mashups may be medleys, but most medleys are probably not mashups. Most medleys keep most of the character of the individual songs and melodies while mashups tend to be more extreme. A mashup is a medley on crank.
