I've tried running both Mame and VMame64 (v0126) on my new PC with Vista Ultimate 64-bit installed. These are the compiled binaries from mamedev.org.
Game speed is great but when I try using the tab-key for configuring ingame options, video and audio start skipping badly and the menus respond very slowly as well.
The same Mame executable works fine on a PC with Vista Home Edition in 32-bit mode.
Is this a known issue?

No issues here.

Thanks for the reply. Can you let me know which version of Vista you're running and which Mame binary (mame, vmame64, etc.) and version?
Also please let me know if you're using the mamedev distribution or if you compiled it yourself.
Appreciate the feedback.

Vista 64 Ultimate SP1
vmame64 (that is the name of the file after it is compiled)
I compiled it myself with the program from headkaze.

Thanks, I'll give that a try. Can you also let me know which mame distribution you're running (ex. 0126, 0127)?
Will report back on whether self-compile solves it.

Solved! :woot
I needed to update my NVIDIA video card drivers to a newer 64-bit version.
Results with Headkaze's compiler were the same as the mamedev binary... but Headkaze's makes it easy to eliminate the warning screens so I'm using that one from now on.

Glad you got it figured out, happy gaming!
