I have not done this so don't take my word for it, but isn't there a "-mouse" command line parameter for MAME?

Yes, you have to enable the mouse controls for use in MAME.  It is just a checkbox in the Controller tab if  you are using MAME32.  If you are using command line MAME, go into the MAME config file and enable the mouse by changing the 0 to 1 in the mouse parameter.  Then save and close the config file and re-launch of refresh MAME.
