Usually, number of buttons isn't the way to go modular. You can play most single, double or triple button games just fine with a six button Street Fighter layout. What you can't do is have a perfect funtioning four way joystick, eight-way, spinners, yoke/steering wheel, etc. all on one panel comfortably.

It's entirely for aesthetics. I just don't like having extra buttons around that don't do anything.
That said, I'm not looking for any absolute authenticity here either. I figure I can make my three button panel sort of arced and add on the single button (already placed lower) and have a decent Neogeo setup with a little fiddling in the input menu. I'll probably do a couple of extra two button panels that line up with the six button for console games, too. Just in case.

Defender , Stargate, and Asteroids used unique button layouts that you might want to build panels for.

I went with a six button layout and then added the thumb button to the joystick panels. You can see an example on my web site below. This works well for just about everything including Asteroids.
I later made some diamond panels for a few special games and also made a special panel for defender/stargate because it is unique. Again pictures on the site.
These cover just about any game I can think of.
