Title: interfacing a megadrive controller to USB

are there any guides, diagrams, drivers ect?

It won't be as easy as just connecting some wires and diodes together like interfacing to a parallel port. You are going to need to program a USB controller chip or else the messy way would be to take a USB gamepad and hack it to use the megadrive controller buttons. However a quick trip to [url]http://www.sealiecomputing.com/retrozone/genesis.html[/url will get you a a preprogrammed USB controller chip and make the project a breeze
