I am thinking about scratch building a dual monitor cabinet for Nintendo games. Before I get too far into this, will Mame let you play games like Punch Out using 2 monitors like the original? If the answer is yes, will Mame also let me use the second monitor to display marquee's when playing a single monitor game like Vs. Super Mario Brothers?

first question is YES
Second is No BUT, you can use MAME hooker or other 3rd party programs to display another image on a second monitor

As Malenko said MAME does indeed support multiple monitors.
as for the marquee thing you'll need an external program, honestly from what I've read the best way to go about this would be to hook into the front end so it sets the marquee before it launches MAME or any other emulator, but I don't have any experience setting this up yet.

Actually.... if you are a glutton for punishment, you can make artwork files that display the marquee on the second screen. Honestly though just install mamehooker use the example marquee program and be done with it.
