Skinnable using HTML5 and Javascript or some simplifying wrapper. Specifically, I'd be after the effects possible using JQuery and HTML5.
I wouldn't necessarily expect a lot of media management options, aside from building/editing playlists. There are already tons of apps out there for managing MP3 tags, etc.
As long as the jukebox read all import tag formats and could utilize the data, that's key.
Also, being able to freely intermix Videos as well as audio, being able to stream vids from the internet (ie Youtube).
Finally, GOOD touchscreen control is essential, single touch is absolutely required, but supporting gestures and multitouch for some elements might be very nice as well.
For instance, JRiver Media Center has a GREAT 10ft presentation, but their touch support and UI fails miserably when actually used on a touchscreen.

The ability to also control the juke from an Android tablet, iPad, or iPhone is also good.
I know for myself I'm using Touch Jams right now, I like it. But I wish the Android side of it looked better.
My issue is the look, I want something polished.

I have nothing more to add other than to say that the software should fully support single touch screens. Make multi-touch an 'option' rather than the 'default.'

Have a singles mode with randomly presented song titles....be a jukebox not a media directory.

Tiny buttons on 'touchscreen' software drives me nuts. Stupid little play and stop buttons when its clearly going to be used by a fat finger makes no sense. Most of the software out is the same.
Edit: Oh and don't put buttons near the edge of screens. e.g. the X in top right. A lot of touchscreens aren't sensitive enough to reach right into the corner with a finger.

If you want a list of what people want you will get more ideas by visiting the jukebox software sites from the list pinned on here and building a list of all their features and geting those down to a list of what you think is important. Once you have all that implimented then get an alpha out. You get far more productive feedback once you have the software out there and people are using it.
There are 3 distinct types of jukebox software for 3 seperate build types. Button Controlled via keypad, Joystick/Arcade controlled and Touch screen controlled. You will probably want to focus on one area first and build up on that later with skins possibly.

+1 on the touch screen support.
I think buttons should be bigger, and maybe not so many. Useless buttons take up room on the screen, making everything else either crowded or having to be made smaller. Either is not good on a touch screen.
Based on this, the juke SW must be able to be easily skinable. Let the user decide what buttons (s)he wants.
Also the little x in the corner to exit? I agree, this is a bad choice for touchscreen. Give me an exit button that I can edit to look the way I want. Also let the button give you the choice to exit to windows desktop or shutdown the PC. (An optional password or number (ref iPhone5) would be nice to allow access to windows desktop.)
