Absolutely, but you'll basically be using it to cut paper rather than shave wood (which in the case of a properly sharpened chisel, actually splits between the cells), so you'll want to sharpen the chisel before and after using it on MDF.

I just finished using using 3/4" and 1/4" chisels to square up all the ends of the dadoes I made in my cabinet sides, and I can tell you they work just fine.

I have Competitions in my 3/4 panel PLUS 1/8" Lexan and they feel fine.  Before you chisel out material, I would try it first in a separate test panel and see how you like the feel.

Of course, you can always add washers as spacers if you do route the material and then want to bring the handles back down shorter.
