If you set up the
action buttons only this way (but the player1 joystick to the arrow keys, and your coin input as keys "5" and "6"), you will have the mame default key setup.
Or you can copy the hotrod layout and use mame's hotrodse setup (For windows mame: "-ctrlr hotrodse" at command line, or "ctrlr hotrodse" in mame.ini)
NeoGeo games would need a special setup. If you use the windows version of mame, v.60 or higher, the new ctrlr .ini files are the way to do this. Just copy the neogeo.ini file in ctrlr/hotrodse folder to ctrlr/
yourcpname and put "ctrlr
yourcpname" in mame.ini.
BTW, you mean "5 1 2 6" as the key layout on the top, yes? By default, "5" = player 1 coin, "6" = player 2 coin.
